The Invention
Building off of their prior discovery (Photo-Cleavable Surfactants P180335US02), UW-Madison researchers disclose an improved method for exosome proteomics. The method leverages the researchers’ photo-cleavable surfactant, Azo, to greatly simplify and improve digestion and protein extraction when compared to current surfactants (e.g., SDS). Exosomes are contacted with Azo in a one-pot preparation, followed by exposure to UV radiation, which degrades Azo leaving the exosome-derived proteins solubilized and ready for subsequent analysis. The researchers successfully analyzed these proteins using mass spectrometry (MS). All told, the present invention coupled with Azo facilitates high-throughput digestion with minimal sample cleanup, resulting in quantitative and highly reproducible characterization of exosome-derived proteins.
